纖毛神經(jīng)營養(yǎng)因子(CNTF)是一種多肽,最初從雛雞胚胎眼組織中純化,并被鑒定為培養(yǎng)中胚胎雛雞纖毛副交感神經(jīng)元的營養(yǎng)因子。隨后的研究表明,CNTF是其他神經(jīng)元細胞類型的存活因子,包括:背根神經(jīng)節(jié)感覺神經(jīng)元,交感神經(jīng)節(jié)神經(jīng)元,胚胎運動神經(jīng)元,主要盆腔神經(jīng)節(jié)神經(jīng)元和海馬神經(jīng)元。CNTF也被證明可以防止軸切開術(shù)后運動軸突的退化。人類CNTF的基因已經(jīng)定位在11號染色體長臂的近端區(qū)域。人類CNTF的cDNA編碼缺乏信號序列的200個氨基酸殘基多肽。CNTF在物種間高度保守,并表現(xiàn)出跨物種活動。人和大鼠CNTF在其蛋白質(zhì)序列中具有約83%的同源性。CNTF在結(jié)構(gòu)上與IL-6,IL-11,LIF和OSM相關(guān)。所有這四種αelix束細胞因子共享gp130作為其受體復合物中的信號轉(zhuǎn)導亞基。
Ciliary neurotrophic factor (CNTF) is a polypeptide initially purified from chick embryo ocular tissue and identified as a trophic factor for embryonic chick ciliary parasympathetic neurons in culture. Subsequent studies have demonstrated that CNTF is a survival factor for additional neuronal cell types including: dorsal root ganglion sensory neurons, sympathetic ganglion neurons, embryonic motor neurons, major pelvic ganglion neurons and hippocampal neurons. CNTF has also been shown to prevent the degeneration of motor axons after axotomy.The gene for human CNTF has been localized to the proximal region of the long arm of chromosome 11. The cDNA for human CNTF encodes a 200 amino acid residue polypeptide that lacks a signal sequence. CNTF is highly conserved across species and exhibits cross-species activities. Human and rat CNTF share approximately 83% homology in their protein sequence. CNTF is structurally related to IL-6, IL-11, LIF and OSM. All of these four αelix bundle cytokines share gp130 as a signal-transducing subunit in their receptor complexes.
